482 Differences in chromatin accessibility in male vs female keratinocytes using ATAC-seq

Sexual dimorphism contributes to autoimmune diseases, such as systemic lupus erythematosus (SLE), where the female-to-male ratio is 9:1. Although hormonal and sex chromosome variation contribute women being more prone predominant driving mechanism(s) remain unknown. We hypothesize that male female keratinocytes display distinct differences in chromatin landscape nucleosome accessibility enforce inflammatory responses. have assessed by performing assay for transposase sequencing (ATAC-seq) on primary (KCs) isolated from healthy donors (n=3, each). In addition, we used ATAC-seq determine changes response pro-inflammatory cytokines (i.e., IL-17A, type I IFN, TNF) KCs compared between female. demonstrate specific sexual vs. KCs. Specifically, our data identifies regions adjacent genes related epidermal differentiation responses responses, particularly IFNs. These corroborate previous ChIP-seq data, showed higher level of H3K4me3 within gene regions. preliminary demonstrates treatment with cytokines, including TNF, leads modification chromosomal accessible KC.
